header_3_font_size=&#8221;25px&#8221; header_3_line_height=&#8221;1.4em&#8221; text_orientation=&#8221;justified&#8221;]<\/p>\n<h3><\/h3>\n<h3>Resum del projecte<\/h3>\n<h3>Els riscs alimentaris associats amb les ciguatoxines: seguiment i supervisi\u00f3 de les toxines marines i organismes productors de toxines als ecosistemes marins (CIGUARISK)<\/h3>\n<p>&nbsp;<\/p>\n<p>La ciguatera \u00e9s una intoxicaci\u00f3 aliment\u00e0ria produ\u00efda pel consum de peixos marins que contenen ciguatoxines (CTX), unes potents neurotoxines principalment presents en les aig\u00fces del tr\u00f2pic i subtr\u00f2pic, i que causen s\u00edmptomes generals, digestius, card\u00edacs i neurol\u00f2gics, aquests poden perdurar durant mesos. Les CTXs s\u00f3n produ\u00efdes per dinoflagel\u00b7lats dels g\u00e8neres Gambierdiscus i Fukuyoa i es transfereixen i transformen a trav\u00e9s de les xarxes tr\u00f2fiques arribant als consumidors.<br \/>Recentment Gambierdiscus spp. ha estat descrit com un component probablement aut\u00f2cton de les comunitats de microalgues bent\u00f2niques a les Illes Can\u00e0ries (IC), la qual cosa suggeriria que aquest g\u00e8nere no \u00e9s un tax\u00f3 recentment introdu\u00eft. El canvi clim\u00e0tic pot contribuir a augmentar les densitats de les poblacions. Des de l&#8217;any 2004 han aparegut diferents casos de ciguatera a les IC i a Madeira.<\/p>\n<p>D&#8217;altra banda, a la Mediterr\u00e0nia, no existeixen proves convincents de casos de ciguatera, si b\u00e9 s&#8217;ha descrit la pres\u00e8ncia de Gambierdiscus spp. a Creta, Xipre i a les Illes Balears (IB). No obstant aix\u00f2, se sap poc sobre la diversitat, distribuci\u00f3 i toxicitat d&#8217;aquests dinoflagel\u00b7lats a la Mediterr\u00e0nia.<br \/>Aquestes dues regions temperades, les IB amb pres\u00e8ncia de Gambierdiscus per\u00f2 sense ciguatera, i les IC amb pres\u00e8ncia de Gambierdiscus i casos de ciguatera, ofereixen una oportunitat \u00fanica per avaluar els factors associats al desenvolupament de la ciguatera, en un entorn que evoluciona com a resultat de l&#8217; canvi clim\u00e0tic.<\/p>\n<p>Hi ha una gran diversitat de CTXs: es coneixen c. 20 an\u00e0legs amb diferents propietats toxicol\u00f2giques. L&#8217;an\u00e0lisi de les CTXs suposa un repte considerable ja que i) l&#8217;efic\u00e0cia dels m\u00e8todes d&#8217;extracci\u00f3 de les CTXs poden variar no nom\u00e9s en funci\u00f3 de l&#8217;esp\u00e8cie de peix sin\u00f3 tamb\u00e9 de el teixit que es analitzi i ii) els nivells de les CTXs observats en mostres de peixos s\u00f3n t\u00edpicament molt baixos.<\/p>\n<p>L&#8217;objectiu principal d&#8217;CIGUARISK \u00e9s fer una avaluaci\u00f3 de el risc de la ciguatera per poder aix\u00ed protegir el consumidor i assessorar els sectors d&#8217;aq\u00fcicultura i pesca. Per aix\u00f2, \u00e9s indispensable determinar l&#8217;abund\u00e0ncia i distribuci\u00f3 espai-temporal de les esp\u00e8cies de Gambierdiscus \/ Fukuyoa, aix\u00ed com la quantitat i composici\u00f3 de les CTXs que produeixen i com aquestes es transformen en les xarxes tr\u00f2fiques. Per aconseguir aix\u00f2, es combinaran mostrejos de camp, cultius i an\u00e0lisis gen\u00e8tics de Gambierdiscus spp. i Fukuyoa spp. amb la caracteritzaci\u00f3 de les CTX caracteritzaran (per exemple, identitat i toxicitat) en els Gambierdiscus spp i en altres components de les xarxes tr\u00f2fiques per veure com s&#8217;acumulen aquestes toxines i com canvien els seus perfils i toxicitats a mesura que es transfereixen al llarg de la cadena tr\u00f2fica (d&#8217;algues a herb\u00edvors \/ omn\u00edvors, i d&#8217;aquests a carn\u00edvors i altres organismes marins). L&#8217;absorci\u00f3 i les conversions de les CTXs en els peixos s&#8217;examinar\u00e0 a trav\u00e9s d&#8217;experiments diet\u00e8tics controlats. Finalment, es construir\u00e0 un model integrat d&#8217;avaluaci\u00f3 dels riscos que predigui quan poden donar-se episodis de ciguatera.<\/p>\n<p>[\/et_pb_text][et_pb_text module_id=&#8221;project&#8221; _builder_version=&#8221;4.9.2&#8243; _module_preset=&#8221;default&#8221; header_text_color=&#8221;#44d89e&#8221; header_3_font=&#8221;|600|||||||&#8221; header_3_text_align=&#8221;center&#8221; header_3_text_color=&#8221;#1bb3eb&#8221; header_3_font_size=&#8221;25px&#8221; header_3_line_height=&#8221;1.4em&#8221; text_orientation=&#8221;justified&#8221;]<\/p>\n<h3>Resum del subprojecte 1<\/h3>\n<h3>La din\u00e0mica espai-temporal de les microalgues productores de ciguatoxines als ecosistemes marins i la transfer\u00e8ncia de ciguatoxines a trav\u00e9s de la xarxa tr\u00f2fica (CIGUATROFIC)<\/h3>\n<p>&nbsp;<\/p>\n<p>La intoxicaci\u00f3 aliment\u00e0ria per ciguatera (CFP), principalment tropical, est\u00e0 causada pel consum de peix que cont\u00e9 ciguatoxines (CTXs). Els dinoflagel\u00b7lats bent\u00f2nics, Gambierdiscus i Fukuyoa produeixen CTXs que s\u00f3n transferides a trav\u00e9s de les xarxes tr\u00f2fiques. El 2004, es va informar sobre el primer brot aut\u00f2cton de ciguatera a les costes europees, exactament a les Illes Can\u00e0ries (IC) despr\u00e9s del consum d\u2019una cerviola (Seriola dumerili). Fins al moment, s&#8217;han descrit cinc esp\u00e8cies de Gambierdiscus a les IC. A la Mediterr\u00e0nia, no s&#8217;han confirmat casos de ciguatera, per\u00f2 s&#8217;ha descrit la pres\u00e8ncia de Gambierdiscus i Fukuyoa a Xipre, Creta i les Illes Balears (IB). El mar Mediterrani, \u00e9s una de les regions m\u00e9s afectades per l&#8217;augment de les temperatures que acompanyen el canvi clim\u00e0tic, el que fa que aquesta regi\u00f3 sigui m\u00e9s adequada per a les esp\u00e8cies tropicals. Per tant, hi ha una necessitat urgent d&#8217;investigar la distribuci\u00f3 espai-temporal de Gambierdiscus i Fukuyoa per definir les possibles ubicacions de risc de ciguatera en les dues \u00e0rees d&#8217;estudi (IC i IB) i estimar l&#8217;escala de temps probable per a la seva aparici\u00f3, i comprendre com es transformen les CTXs a trav\u00e9s de les xarxes tr\u00f2fiques. Les comparacions entre IC i IB s\u00f3n particularment informatives de el risc de ciguatera ja que els organismes productors de CTXs s\u00f3n presents en ambdues \u00e0rees, per\u00f2 la ciguatera actualment nom\u00e9s s&#8217;ha detectat en CI.<\/p>\n<p>La pres\u00e8ncia de CTXs en la carn de peixos de nivell tr\u00f2fic superior, particularment un dels an\u00e0legs m\u00e9s t\u00f2xics (CTX-1B), s&#8217;explica com a resultat de el consum de Gambierdiscus per peixos herb\u00edvors i peixos omn\u00edvors que al seu torn s\u00f3n consumits per peixos carn\u00edvors . El subp. 1, CIGUATROFIC, se centra en on i quan s\u00f3n presents els dinoflagel\u00b7lats productors de CTXs, qu\u00e8 controla el seu creixement, els seus perfils de CTXs i com aquests perfils canvien a trav\u00e9s de les xarxes tr\u00f2fiques. Les distribucions espaials de Gambierdiscus i Fukuyoa a IB i IC s&#8217;estudiaran i quantificaran mitjan\u00e7ant recomptes microsc\u00f2pics i t\u00e8cniques moleculars (seq\u00fcenciaci\u00f3 Sanger i ADN metabarconding). La identificaci\u00f3 a nivell molecular \u00e9s necess\u00e0ria ja que la toxicitat de les esp\u00e8cies de Gambierdiscus varia considerablement i la distinci\u00f3 a nivell d&#8217;esp\u00e8cie a partir de la morfologia no \u00e9s possible o de el tot fiable. En alguns llocs clau, on les abund\u00e0ncies de Gambierdiscus siguin elevades, s&#8217;han de determinar tamb\u00e9 les distribucions temporals i en profunditat.<\/p>\n<p>Les CTXs produ\u00efdes pels dinoflagel\u00b7lats es caracteritzaran i compararan amb les presents en els peixos i altres organismes per determinar com canvia la composici\u00f3 i la toxicitat de les CTXs durant el seu pas a trav\u00e9s de les xarxes tr\u00f2fiques. Les CTXs s&#8217;analitzaran mitjan\u00e7ant assajos cel\u00b7lulars i cromatografia l\u00edquida acoblada a espectrometria de masses (LC-MS) per confirmar la pres\u00e8ncia, identificaci\u00f3 dels an\u00e0legs de CTXs i les quantitats de cada CTX present. Aquesta caracteritzaci\u00f3 de les CTXs en els diferents nivells de la xarxa tr\u00f2fica ajudar\u00e0 a comprendre la transformaci\u00f3 i la biomagnificaci\u00f3 de les toxines i la seva toxicitat en la xarxa tr\u00f2fica. Can\u00e0ries \u00e9s una de les regions on es constata un notable increment de la pres\u00e8ncia de ciguatoxina i altres derivats an\u00e0legs en peixos i casos d&#8217;intoxicacions humanes de ciguatera, pel que representa un magn\u00edfic laboratori per centrar aquest estudi. S\u00f3n diverses les toxines que pertanyen a aquesta fam\u00edlia i poden vehicular-les fins al consumidor. D&#8217;altra banda, aquestes toxines produeixen un efecte bioacumulatiu a la cadena tr\u00f2fica, de qu\u00e8 es desconeix el seu mecanisme prec\u00eds, la manera en qu\u00e8 afecta els perfils tox\u00ednics, i com aquests perfils podrien influir en la seva capacitat t\u00f2xica en cada una de les baules de la cadena.<\/p>\n<p>En aquest subprojecte pretenem obtenir nous cultius de Gambierdiscus, a\u00efllant de mostrejos rutinaris en punts concrets (hotspots) per a la seva incorporaci\u00f3 al Banc Espanyol d\u2019Algues (BEA) despr\u00e9s de la seva identificaci\u00f3 morfol\u00f2gica i gen\u00e8tica, aix\u00ed com millorar la taxa de creixement i producci\u00f3 de biomassa d\u2019aquesta microalga, principal handicap contra l&#8217;avan\u00e7 del coneixement de la participaci\u00f3 d&#8217;aquest g\u00e8nere com a responsable inicial de les intoxicacions en humans. Amb aquests cultius es realitzar\u00e0 un estudi dels seus perfils tox\u00ednics, i la que es seleccioni en aquesta fase es far\u00e0 servir en els experiments de bioacumulaci\u00f3.<\/p>\n<p>D&#8217;altra banda, els possibles organismes vectors de la ciguatera podrien ser diversos, i a hores d&#8217;ara desconeguts amb exactitud, al no existir estudis que demostrin la proced\u00e8ncia exacta d&#8217;aquestes esp\u00e8cies i la correspond\u00e8ncia amb els vectors de transmissi\u00f3 d&#8217;aquesta malaltia. En aquest projecte pretenem determinar les possibles esp\u00e8cies que actuen com a vectors de la ciguatera en els diferents nivells tr\u00f2fics, analitzant peixos i equinoderms recollits en els mateixos punts descrits en l&#8217;apartat anterior (hotspots), que a causa de la seva alimentaci\u00f3 a base d&#8217;algues, ja sigui de forma exclusiva com a \u00fanica font d&#8217;alimentaci\u00f3, o associada amb petits peixos i \/ o crustacis, siguin susceptibles d&#8217;acumular ciguatoxina, per posteriorment investigar altres esp\u00e8cies que actuen en altres nivells de la cadena aliment\u00e0ria, arribant als grans predadors, principals responsables de les intoxicacions en l&#8217;esp\u00e8cie humana.<\/p>\n<p>En aquest subprojecte tamb\u00e9 pretenem estudiar els efectes de la bioacumulaci\u00f3 d&#8217;aquestes toxines en els peixos, mitjan\u00e7ant assajos d&#8217;exposici\u00f3 a la toxina en animals experimentals, utilitzant la seva alimentaci\u00f3 amb cultius de microalgues t\u00f2xiques seleccionades durant el projecte i, paral\u00b7lelament, alimentant-los amb carn de peix pr\u00e8viament identificada i quantificada amb una quantitat (ng\/g) de ciguatoxina. Aquests assajos ens ajudaran a con\u00e8ixer millor la cin\u00e8tica de la bioacumulaci\u00f3 de les toxines en els peixos. (CIGUARISK)<\/h3>\r\n\u00a0\r\n\r\nCiguatera, also known as ciguatera fish poisoning (CFP), is caused by the ingestion of marine fish containing ciguatoxins (CTXs), which are very potent neurotoxins, mainly present in tropical and subtropical areas worldwide. Ciguatera causes general, digestive, cardiac and long-lasting neurological symptoms that may last for months. CTXs are produced by dinoflagellates of the genera Gambierdiscus and Fukuyoa and are transferred and transformed through the food webs reaching consumers.\r\n\r\nGambierdiscus spp. have been described as a widespread and diverse component of the benthic microalgal communities in the Canary Islands (CI). It is unlikely that the genus is a recently introduced taxon in the Canaries but climate change may have promoted higher population densities. Since 2004, cases of ciguatera have appeared in the CI and in Madeira. \r\n\r\nBy contrast, in the Mediterranean Sea, no convincing evidence of cases of ciguatera exist, even though Gambierdiscus spp. have been reported in Crete, Cyprus and very recently in the Balearic Islands (BI). However, little is known about the diversity, distribution and toxicity of Gambierdiscus and Fukuyoa in the Mediterranean Sea. \r\n\r\nThese two temperate regions \u2013 the BI having Gambierdiscus but no ciguatera, the CI having both \u2013 offer a unique opportunity to evaluate the factors that allow a potential risk of ciguatera to become an actual disease and hence to develop risk assessments for ciguatera in an environment evolving as a result of climate change.\r\n\r\nThe toxins involved are very diverse: c. 20 CTXs analogues are known, with very different toxicological properties. Analysing them is challenging since extraction methods for these toxins may not be equally efficient in different species of fish and different tissues, and the levels of CTXs observed in fish samples are typically very low. \r\n\r\nThe main objective of CIGUARISK is to make an integrated assessment of ciguatera risk in order to protect the consumer and advise the fish production sector. A basic prerequisite is to determine the spatiotemporal distribution of Gambierdiscus\/Fukuyoa species, precisely identify and quantify the CTXs they produce, and define how CTX profiles are transformed during their passage through the food webs. To accomplish this, field sampling and DNA profiling of Gambierdiscus spp. and Fukuyoa spp. will be combined with CTX characterization (identity and toxicity) in the dinoflagellates and various components of food webs to see how CTX accumulate and how their profiles and toxicities change as they are transferred from alga to herbivorous\/omnivorous and to carnivorous fish and other marine organisms. Controlled dietary experiments will be performed to examine uptake and conversions of CTXs by fish. Benthic dinoflagellates of the genera Gambierdiscus and Fukuyoa produce CTXs, which are then transferred up the food web. In 2004 the first autochthonous outbreak of ciguatera in Europe was reported from the Canary Islands (CI) after consumption of amberjack fish. Until now, five Gambierdiscus species have been reported in the CI. In the Mediterranean, no ciguatera cases have been confirmed, but Gambierdiscus and Fukuyoa species have been found in Cyprus, Crete and the Balearic Islands (BI). The Mediterranean Sea is strongly affected by rising temperatures accompanying climate change, making the region increasingly suitable for tropical species. Hence there is an urgent need to investigate the spatio-temporal distributions of Gambierdiscus and Fukuyoa to define locations where ciguatera is a potential risk and estimate the likely timescale for its appearance, and to understand how CTXs move through the food web. Comparisons between CI and BI are particularly informative about ciguatera risk because CTX-producers are present in both areas but ciguatera currently develops only in CI.\r\n\r\nThe presence of CTXs in the flesh of upper trophic level fish, particularly one of the most toxic analogues (CTX-1B), is believed to result from consumption of Gambierdiscus cells by herbivorous and omnivorous fish; then, these are preyed upon by carnivorous species. Subproject 1 CIGUATROFIC focuses on where and when CTX-producing dinoflagellates occur, what controls their growth, their CTX profiles, and how these profiles change as CTXs are transferred in the food webs. The spatial distributions of Gambierdiscus\/Fukuyoa in BI and CI will be studied and quantified by microscopical counts, and molecular techniques (Sanger sequencing and DNA metabarcoding). Molecular approaches for identification are needed because CTX-producing species vary greatly in toxicity but cannot be reliably identified by morphology. Temporal and depth distributions will also be examined at a few selected sites where CTX species are sufficiently abundant.\r\n\r\nThe CTXs produced by the dinoflagellates will be characterized and compared with those present in fish and other organisms to determine how CTX composition and toxicity change during their passage through the food webs. Previous studies indicate that CTXs become more oxidized (e.g. to CTX-1B) as they move through the food web and accumulate in fish tissues because of their lipophilic nature. Screening of CTXs will employ cell-based toxicity assays and liquid chromatography coupled to mass spectrometry (LC-MS\/MS and LC-HRMS) to confirm the presence and identities of CTX analogues and their amounts. This characterization of the CTXs in different organisms will help to understand the biotransformation and biomagnification of toxins and toxicity in the food web. Bioaccumulation of CTXs is usually evaluated in muscle tissue but we will also evaluate it in other tissues (liver, stomach) to provide a more complete picture. The information from the spatiotemporal studies of dinoflagellates and toxin analyses in marine organisms will then be integrated to develop a predictive model for ciguatera.[\/et_pb_text][et_pb_text module_id=\"subproject2\" _builder_version=\"4.7.6\" _module_preset=\"default\" header_text_color=\"#44d89e\" header_3_font=\"|600|||||||\" header_3_text_align=\"center\" header_3_text_color=\"#1bb3eb\" header_3_font_size=\"25px\" header_3_line_height=\"1.4em\"]<h3>SubProject 2 summary<\/h3>\r\n<h3>Ciguatoxin in food web: a controlled study of bioaccumulation in fish species (CIGUAFOOD)<\/h3>\r\n<p>\u00a0<\/p>\r\n<p>Emergent marine toxins are a major problem involving food safety for fishing and aquaculture products in the world. Marked increase of the prevalence of ciguatoxin and other analogue derivatives in fish and ciguatera fish poisoning in humans has been detected in different regions like the Canary Islands, which represents a best lab to address this issue. Different toxins belong this family could be transmitted to the consumer. And these toxins produce a bio-accumulative effect in the food web, but nothing is known about their mechanism of action, how it affects the toxins profiles, and their influence on the toxic activity for each point of this food web.<\/p>\r\n<p>New strains of <em>Gambierdiscus<\/em> will be obtained in this subproject from the hotspots described in Gran Canaria, in order to be included in the Spanish Algae Bank (BEA) collection after their morphologic and genetic identification, but also the rate of growth and biomass production improved. This knowledge is necessary to clarify their contribution in the human intoxication outbreaks by these toxins. These isolated strains will be used to deepening in the toxinic profiles found, and that showing the best results will be included in the bioaccumulation study.<\/p>\r\n<p>In the other way, different organisms could participate as carriers for ciguatera fish poisoning. Possible species acting as ciguatera fish poisoning carriers in the different points of the food web will also be studied in this project, and checked in the same points described above (hotspots), by using algae as main source of food, or mixed with small fish and shellfish, and both groups are able to accumulate ciguatoxin and other toxin derivatives involved, and finally, main fish predators will also be investigated for their responsibility in human ciguatera fish poisoning.<\/p>\r\n<p>And the effect of bioaccumulation of these toxins in fish will be also studied in this project, by different challenge exposure to the toxins in experimental fish, by feeding with microalgae strains selected during this project and, at the same time, with death fish, previously identified and quantified for a particular amount (ng\/g) of ciguatoxin.
